Nintendo releases a new home system every five years without fail since the Super NES: SNES in 91, N64 in 96, GCN in 01 and Wii in 06. Therefore, it would be logical to predict a 2011 release for the new console and an unveiling at this year's E3.
However, things are different this time around; sales of the Wii are still phenomonal, there doesn't seem to be any let up in third party support and the Xbox360 and PS3 have both been marketted to last longer than previous consoles.
Therefore, I'd tip the release date to be closer to 2013. Which, yes, is after the end of the world. Looks like the Wii will be the last console we see from Nintendo.

Ok, what is it with people and the end of the world these days? This is shit, because we've survived Y2K, Mad Cow, Bird Flu, and Swine Flu. I remember there was a rumor by the time 2000 came around, we would somehow not exist. And then there's Ice Age theories, and global warming and global cooling contradict each other. So, seriously, this is not about to happen. Just because the Mayan Calendar ended at December 21, 2012 doesn't mean anything. THEY HAVE LIVES, TOO, YOU KNOW! They probably had some other stuff to do, like take care of their tribe and children. Now if you were just kidding, nice job on getting me started.
Anyway, by that time, there will also be a new Xbox 720, and possibly a PS4.
